Atomic Force Microscopy (AFM) as one of the cutting-edge nanotechnology has developed fast in recent years. The most recent commerial product has realized vedio-rate visualization of biomolecular processing in nanoscale. Herein, AFM is used to thread a tethered peptide chain through a subnanopore so that the translocation is systematically controlled, which can be the funfamentals in the next-generation sequencing development. In addition, capabilities of nanoindentation, resonance frequence shift weighing, conductive AFM, force spectroscopy and bio-compatible imaging, makes AFM promising in many interdisciplinary fields such as synthetic biology and virology.
