单相炭材料的生物相容性

Abstract:对树脂炭、炭纤维、热解炭和石墨4种单相炭材料在模拟体液中进行材料表面颗粒脱落以及生物相容性的研究。采用模拟体液震荡浸泡法研究材料表面颗粒脱落量;用噻唑蓝法和直接接触实验研究材料的生物相容性,用倒置生物显微镜对细胞的生长形貌进行观察。结果表明:在模拟体液中浸泡72 h时4种材料的表面颗粒脱落量均达到峰值,随浸泡时间继续延长,颗粒的脱落量下降并趋于平稳。在168 h内脱落颗粒的总量以石墨最大,为3.01 mg,树脂炭最小,为2.23 mg。4种单相炭材料浸提液接触的细胞在168 h内相对增殖率(relative growth rate, RGR值)均大于75%,细胞毒性等级均为1级。4种单相炭材料均会抑制细胞的生长与增殖,抑制程度由大到小依次为石墨、热解炭、炭纤维和树脂炭。单相炭材料对细胞的毒性机理为:炭颗粒进入细胞造成线粒体降解,使细胞的能量站受到破坏,损伤细胞的DNA,从而导致细胞畸形生长甚至死亡
