Abstract:Differentiation requires coordinated regulators including TET (ten-eleven translocation) enzymes, which oxidize 5-methylcytosine in DNA. Using Keratin 14-Cre-driven ablation of Tet genes in skin epithelial cells, loss of Tet2/Tet3 alters hair shape and length and causes hair loss. Through DNA demethylation, Tet2/Tet3 control chromatin accessibility, Dlx3 binding and promoter activity of Krt25 and Krt28 and their enhancer interactions. Tet2/Tet3 also control 3D chromatin topology at keratin type I/II loci independently of DNA methylation, implicating DNA methylation in hair growth control.
