Abstract:IgNAR shows promise in cancer and antiviral biotherapies. Its variable region (VNAR) binds antigen as strongly as IgNAR but weighs only ~12 kDa, yet antigen-specific VNAR screening remains challenging: phage display suffers from limited library coverage, low expression, unstable target protein and time-consuming procedures. Using next-generation sequencing (NGS) we identified ~3000 BAFF-specific VNARs in Chiloscyllium plagiosum vaccinated with BAFF; modelling, molecular dynamics and ELISA showed that one of the top five abundant VNARs bound BAFF more strongly than those from phage display.
