Abstract:Antibodies play crucial roles in disease diagnosis and therapy. Shark-derived nanobodies (VNARs) are the smallest known antigen-binding fragments (~12 kDa), allowing efficient antigen targeting. Traditional phage display for screening shark nanobodies is limited, as not all nanobodies express well in this format. We used Cre recombinase as antigen, integrating next-generation sequencing, cell sorting, phage display and mass spectrometry; diverse strategies yielded many VNARs, and assays in mammalian cells confirmed specific binding to Cre without inhibiting its activity.
