Abstract:Epigenetics is pivotal in gene expression regulation and cellular differentiation. DNA methylation is a well-known epigenetic mark, and the discovery of 5-hydroxymethylcytosine (5hmC) has given new insight into cytosine modifications. 5hmC, oxidized from 5-methylcytosine (5mC), acts both as an intermediate of demethylation and as a stable genomic modification. This review summarizes the functions of 5hmC in development and disease, its potential as a diagnostic and prognostic marker, and the latest methods for detecting and quantifying 5hmC.
